Acknowledgments
Phil Jackson and Manuel Gurule helped generate data proving that vitamin
C, octylamine, and dithiothreitol uncouples (and dimedone enhances) the
down-regulation of HMG-CoA reductase by an endogenous monooxygenase. Although
deduced by Dr. Knight prior to employment at the University of New Mexico,
his studies proving this mechanism for the regulation of HMG-CoA reductase
were performed in the laboratories of Dr. Terence Scallen at UNM under
NIH Grants HL16796 and AM10628. Funding from Sandoz Pharmaceuticals for
the verification and large-scale application of Dr. Knight's improvements
to existing assays for HMG-CoA reductase is also gratefully acknowledged.
In addition to helping to resolve and confirm the monooxygenase and reductase
regulatory couple, these assays were used to screen a large number of prospective
inhibitors for HMG-CoA reductase which eventually led to the identification
and FDA approval of Lescol (®Sandoz Pharmaceuticals) for use as a cholesterol-lowering
drug.
